The biochemist and geneticist David H MacLennan, OC, OOnt, FRSC, FRS (born July 3, 1937) is best known for his fundamental research on proteins that regulate calcium flux through the sarcoplasmic reticulum (SR), thereby controlling muscle contraction and relaxation, as well as for his discoveries in the field of muscle diseases caused by genetic defects in calcium regulatory proteins. He was born in Swan River, Manitoba, and graduated with a BSA from the University of Manitoba in 1959, followed by a DSc (hc) in 2001 from the same institution. In 1961, he got his MS and PhD from Purdue University, after which he spent three years as a Postdoctoral Fellow (1963-1964) and three years as an Assistant Professor (1964-1968) at the University of Wisconsin.
Associate Professor at the Banting and Best Department of Medical Research in 1969, he was promoted to Professor in 1974, Chair in 1978, J. W. Billes Professor of Medical Research in 1987, and finally University Professor in 199(1993-). He has made significant advances to our knowledge of the mechanism of ion transport by sodium-dependent calcium pumps, the storage and release of calcium from the SR by acidic lumenal proteins, and the transport of calcium from the SR by calcium release channels. A team he headed was responsible for defining the genetic foundation for human skeletal muscle illnesses such as malignant hyperthermia and central core disease, as well as Brody disease. He has also proven that mutations in phospholamban, a regulator of the calcium pump, may result in cardiomyopathy. His discovery of a calcium release channel mutation that causes porcine stress syndrome led in the development of a diagnostic test that has resulted in a significant reduction in the incidence of the illness, resulting in significant economic advantages for the pig business.
